چکیده

A NATO-sponsored Advanced Research Workshop (ARW 990-83) on "Coordinate Regulation of Cholesterol Metabolism" was held from October 1-3, 1984, in Sante Fe, New Mexico. The Giovanni Lorenzini Foundation of Italy was also a sponsor. The organizing director was Dr. Ajit Sanghvi, University of Pittsburgh; the co-directors were Drs. Giovanni Galli, University of Milan, Italy, and Terence J. Scallen, University of New Mexico. Drs. David Kritchevsky, The Wistar Institute, Philadelphia, and George Vahouny, George Washington University, Washington, D.C., served on the advisory committee. The last 10 years have provided considerable information on the regulation of cholesterol metabolism via lipoprotein receptor action in various types of cells. A significant body of information about in vitro changes in the activities of cholesterol-metabolizing enzymes has also developed. Changes in the state of phosphorylation of HMG-CoA reductase by experimental manipulations has been shown to be correlated with changes in the enzyme activity. The experimental systems used in these studies have used broken-cell preparation, intact heptatocytes, and whole animals. Similar but less extensive information has recently been gathered for acyl-coA:cholesterol acytransferase (ACAT) and cholesterol 7a-hydroxylase, the other two major enzymes responsible for cholesterol metabolism in the liver cell. There is debate as to whether phosphorylation/dephosphorylation of these enzymes plays a role in vivo and, if it does, the extent of its significance. However, those who suggest enzyme phosphorylation/dephosphorylation as a possible in vivo mechanism for regulating cholesterol flow within the cell suggest it as only a short-term regulatory phenomenon; changes in protein synthesis are required for the long-term.

Bile acid regulation of gene expression: roles of nuclear hormone receptors.

Bile acids derived from cholesterol and oxysterols derived from cholesterol and bile acid synthesis pathways are signaling molecules that regulate cholesterol homeostasis in mammals. Many nuclear receptors play pivotal roles in the regulation of bile acid and cholesterol metabolism. Bile acids activate the farnesoid X receptor (FXR) to inhibit transcription of the gene for cholesterol 7alpha-hy...

MicroRNAs: emerging roles in lipid and lipoprotein metabolism.

PURPOSE OF REVIEW MicroRNAs (miRNAs) regulate gene expression by binding to target mRNAs and control a wide range of biological functions. Recent reports have identified specific miRNAs as major regulators of fatty acid and cholesterol homeostasis. This review examines the biological function of various miRNAs and the emerging evidence linking specific miRNAs to critical pathways in lipid metab...

In vivo and in vitro studies on the regulatory link between 3-hydroxy-3-methylglutaryl coenzyme A reductase and cholesterol 7 alpha-hydroxylase in rat liver.

The activities of 3-hydroxy-3-methylglutaryl CoA reductase (HMGCoA reductase; EC 1.1.1.34), rate-limiting enzyme of cholesterol biosynthesis, and cholesterol 7 alpha-hydroxylase (EC 1.14.13.17), key enzyme of the neutral bile acid synthesis pathway, were measured in the microsomal fraction of rat liver and in rat liver cells to investigate the coordinate regulation of the two pathways. Both enz...

Identification of the ER-resident E3 ubiquitin ligase RNF145 as a novel LXR-regulated gene

Cellular cholesterol metabolism is subject to tight regulation to maintain adequate levels of this central lipid molecule. Herein, the sterol-responsive Liver X Receptors (LXRs) play an important role owing to their ability to reduce cellular cholesterol load. In this context, identifying the full set of LXR-regulated genes will contribute to our understanding of their role in cholesterol metab...

SUMMARY Due to the importance of coronary heart diseases in today's world and in view of the fact that hyperlipidemia, hyperglycemia and obesity are regarded as the main risk factors in these diseases and also considering the paucity of epidemiologic surveys in our society, this study was carried out in Islamshahr 30 Km to the southwest of Tehran. Out of 244000 population of the area, 2033 per...
